Novel preclinical models

CXR have developed innovative preclinical screening models to improve lead compound selection by providing a better understanding of metabolism and toxicity.

  • transADMET™ Mice. CXR have replaced murine PXR and CAR by human genes, and these humanised mice reflect known species differences in drug disposition and toxicity.
  • The HRN™ mice, that lack hepatic P450 metabolism.
  • Tox Reporter mice, containing engineered biomarkers that report on mechanism of toxicity.
  • Microsampling PK studies: all of these models (as well as wild types) are amenable to our microsampling PK sampling procedure.

Mechanistic assays and screens

CXR Biosciences have developed a range of mechanistic assays (examples below) and can rapidly develop bespoke assays and screens tailored to the client's requirements

  • The AREc32 cell line, an in vitro screen for electrophilic and antioxidant properties of chemicals, and potential in vitro alternative to the Local Lymph Node Assay.
  • Tox Reporter cell lines; cell-based reporter systems that report on specific mechanisms of toxicity of chemicals and drugs.
  • Transactivation assays; cell based transactivation assays that report on the interaction of compounds with nuclear hormone receptors from a variety of species.

Microarray Services

CXR Biosciences employs its microarray services to provide a detailed understanding of the cellular responses to compounds. This is useful in a variety of ADMET applications, but especially in Investigative Toxicology where it is thoughtfully applied to generate hypotheses about the mechanism of toxicity.
